地被菊β微管蛋白基因的克隆及表达分析 Cloning and expression profile analysis of CmTUB gene of ground-cover chrysanthemum 发布时间:2011-06-13 浏览量:84 收藏数:0 评论数:0 总览 评价 夏胜军1, 刘兆磊2, 蒋甲福2, 陈素梅2, 张淑梅2, 陈发棣2* ( 1、南京农业大学园艺学院; 2、南京农业大学园艺学院,南京 210095; ) 摘要: 通过RT-PCR、RACE技术从地被菊'雨花金华'中成功克隆了一个β微管蛋白基因,命名为CmTUB,基因登录号为AB608732。序列分析发现,CmTUB基因cDNA全长为1544bp,编码446个氨基酸,氨基酸序列由一个包含198个氨基酸残基的GTPase结构域和138个氨基酸残基的C-端组成,并包含6个与微管蛋白连接相关的功能域。系统进化树分析表明与棉花等双子叶植物亲缘关系较近。荧光定量PCR分析结果表明,CmTUB基因属于组成型表达基因,重力可以抑制CmTUB在背地侧的表达;NAA和CaCl2可以促进涂抹侧的表达;TIBA和EGTA则对涂抹侧CmTUB的表达具有抑制作用。 关键词: 地被菊;β微管蛋白;序列分析;实时荧光定量 XIA Shengjun1, LIU Zhaolei2, JIANG Jiafu2, CHEN Sumei2, ZHANG Shumei2, CHEN Fadi2* ( 1、Horticulture college of Nanjing agriculture university; 2、Horticulture college of Nanjing agriculture university, Nanjing 210095; ) Abstract: A β-tubulin gene named CmTUB was successfully cloned from chrysanthemum 'Yuhuajinhua' by RT-PCR and RACE technology, the gene accession number is AB608732. Sequence analysis showed that cDNA full length of chrysanthemum 'Yuhuajinhua' CmTUB gene is 1544bp, encoding 446 amino acids, which consists of 198 amino acid residues of the GTPase domain and 138 amino acid residues of the C-terminal component and includes six connection with tubulin domain-related. Phylogenetic analysis showed that cotton and other dicots have closer relationship with chrysanthemum 'Yuhuajinhua' CmTUB gene. Quantitative PCR analysis showed that CmTUB gene is constitutively expressed genes, gravity can inhibit the expression of CmTUB in the upper side; NAA and CaCl2 can promote the expression of CmTUB in the coated side; TIBA and EGTA can inhibit the expression of CmTUB in the coated side.
